Onion Skin

Study of Onion Skin Cells and Osmosis

Materials needed: one small piece on an onion layer, Tweezers, table salt, a small cup for mixing salt and water, one Blister slide, a Blister Microscope.

1. Cut a piece of the onion layer small enough to fit in the Blister slide.

2. Using the tweezers, peel the thinnest possible layer of onion skin from the onion layer. (see photo)

3. Gently lay the skin as flat as possible in the bottom of the Blister slide. Moistening the skin with a few drops of water may make it easier to flatten.

4. Observe the skin through the Blister Microscope with all objectives, starting with the lowest power objective first (25x, then 50x, then 100x).
5. Prepare some salty water by mixing about ¼ to ½ teaspoon of salt with about 1/8 cup of warm water.

6. Remove the Blister from the microscope and pour enough salt water into the blister to cover the onion skin and let it stand for 15 or 20 minutes.

7. Put the blister back under the microscope and observe the onion skin in the salt water through all objectives.

8. Observe any differences in the appearance of the cells after their exposure to the salt water.
